It has been suggested that biogeographic historical legacies in plant diversity may influence ecosystem functioning. This is expected because of known diversity effects on ecosystem functions, and impacts of historical events such as past climatic changes on plant diversity. However, empirical evidence for a link between biogeographic history and present‐day ecosystem functioning is still limited. Here, we explored the relationships between Late‐Quaternary climate instability, species‐pool size, local species and functional diversity, and the net primary productivity (NPP) of Northern Hemisphere forests using structural equation modelling. Our study confirms that past climate instability has negative effects on plant functional diversity and through that on NPP, after controlling for present‐day climate, soil conditions, stand biomass and age. We conclude that global models of terrestrial plant productivity need to consider the biogeographical context to improve predictions of plant productivity and feedbacks with the climate system.  相似文献

Temporal evolution of dissolved and biogenic silica concentrations along the Scheldt tidal river and in its tributaries was investigated during 1 year in 2003. In the tributaries, dissolved silica (DSi) concentrations remained high and biogenic silica (BSi) concentrations were low throughout the year. In the tidal river during summer, DSi was completely consumed and BSi concentrations increased. Overall, most of the BSi was associated with living diatoms during the productive period in the tidal river. Nevertheless, the detrital BSi was a significant fraction of the total BSi pool, of which less than 10% could be attributed to phytoliths. The tidal river was divided into two zones for budgeting purposes. The highest productivity was observed in the zone that received the highest water discharge, as higher riverine DSi input fluxes induced presumably a less restrictive DSi limitation, but the discharge pattern could not explain all by itself the variations in DSi consumption. Silica uptake and retention in the tidal river were important at the seasonal time-scale: from May to September, 48% of the riverine DSi was consumed and 65% of the produced BSi was deposited, leading to a silica (DSi + BSi) retention in the tidal river of 30%. However, when annual fluxes were considered, DSi uptake in the tidal river amounted to 14% of the DSi inputs and only 6% of the riverine silica (DSi + BSi) was retained in the tidal river.  相似文献

A method based on solid-phase extraction (SPE) and capillary zone electrophoresis (CZE) for the analysis of tobramycin in human serum is presented. An off-line SPE employing a carboxypropyl bonded phase (CBA) cartridge was used for the extraction of tobramycin from human serum. Adsorbed tobramycin was eluted from the CBA cartridge using a mixture of NH(3) (25%, w/v)-methanol (30:70, v/v). After evaporation, the analyte was reconstituted and derivatized with o-phthaldialdehyde (OPA)/3-mercaptopropionic acid (MPA). The resulting tobramycin-OPA/MPA derivative was purified, and then identified by mass spectrometry. The tobramycin-OPA/MPA derivative was then analysed by CZE with a background electrolyte (BGE) comprising of 30 mM sodium tetraborate pH 10.0-acetonitrile (ACN) (80:20, v/v) with ultraviolet detection at 230 nm. A linear response was observed in the range of 0.3-30 microg/ml with r(2) = 0.992. The sensitivity of the method was determined by its limit of quantitation (LOQ) and limit of detection (LOD) of 0.3 microg/ml and 0.1 microg/ml, respectively. SPE recovery ranged from 68 to 79% at the trough levels to 98% at the peak levels found in serum. Furosemide has been added as internal standard (IS) to improve precision. For the therapeutic range of tobramycin in serum (2-10 microg/ml) the relative standard deviation (R.S.D.) was less than 11% for the entire SPE/CE process. The method demonstrated excellent selectivity as shown by the lack of interference from a total of 20 drugs investigated. The method was then used in therapeutic drug monitoring of patients receiving the drug.  相似文献

Disruption of leptin signaling in the heart may contribute to obesity-related cardiac disease, as leptin deficient (oblob) mice display cardiac hypertrophy, increased cardiac apoptosis and reduced survival. Since leptin maintains a tonic level of neuronal nitric oxide synthase (NOS1) expression in the brain, we hypothesized that leptin deficiency would decrease NOS1 cardiac expression, in turn activating xanthine oxidoreductase (XOR) and creating nitroso-redox imbalance. We studied 2- to 6-month-old oblob (n=26) and C57Bl/6 controls (n=27). Cardiac NOS1 protein abundance (P<0.01) and mRNA expression (P=0.03) were reduced in oblob (n=10 and 6, respectively), while NOS3 protein abundance and mRNA expression were unaltered. Importantly, cardiac NOS1 protein abundance was restored towards normal in oblob mice after leptin treatment (n=3; P<0.05 vs leptin untreated oblob mice). NO metabolite (nitrite and nitrate) production within the myocardium was also reduced in oblob mice (n=5; P=0.02). Furthermore, oxidative stress was increased in oblob mice as GSH/GSSG ratio was decreased (n=4; P=0.02). Whereas XOR activity measured by Amplex Red fluorescence was increased (n=8; P=0.04), XOR and NADPH oxidase subunits protein abundance were not changed in oblob mice (n=6). Leptin deficiency did not disrupt NOS1 subcellular localization, as NOS1 co-localized with ryanodine receptor but not with caveolin-3. In conclusion, leptin deficiency is linked to decreased cardiac expression of NOS1 and NO production, with a concomitant increase in XOR activity and oxidative stress, resulting in nitroso-redox imbalance. These data offer novel insights into potential mechanisms of myocardial dysfunction in obesity.  相似文献

State‐of‐the‐art technologies that implement theindustrial ecology concept only make it to the market if environmental gains and economic benefits are significant. Therefore, the article investigates, in an interdisciplinary way, two innovative technologies that valorize stainless steel (SS) slags as block masonry (bricks): carbonation and thermo‐alkali‐activation. The technical, environmental, and economic features of three SS bricks—solid bricks, perforated bricks, and lightweight aerated blocks—are compared to commercially available construction materials. Although the produced bricks meet industrial standards, technical challenges, such as optimization of alkali addition and use of metal molds, should be dealt with before upscaling to industrial production. A cradle‐to‐gate life cycle assessment that aggregates the results of the various impact categories shows that the environmental impact of solid and perforated SS bricks is lower than the impact of conventional clay‐baked bricks owing to the avoidance of additives for slag stabilization and energy consumption for sintering clay. The impact of aerated SS bricks was found to be similar to the commercially available aerated blocks. More specifically, the carbon dioxide uptake from carbonation reduces the overall environmental impact, whereas use of alkalis increases the impact. A strengths weaknesses opportunity threats analysis highlights the economic advantages of SS bricks originating from lower energy requirements, reduced dependence on primary resources, and improved metal recovery from slag. However, in order to apply the innovative technologies at industrial scale, challenges related to processing conditions, feedstock variability, and potential competition from existing brick suppliers have to be overcome.  相似文献

To evaluate the effect of short-term changes in discharge on plankton dynamics, a station in the upper reaches of the Schelde estuary was monitored during 1 month in spring 1998 with a sampling frequency of 1–2 days. During this monitoring period, a flood event occurred during which discharge increased about 5-fold. This flood event had a strong effect on the composition of the planktonic community. Rotifers were strongly negatively affected by the flood event while densities of heterotrophic nanoflagellates and scuticociliates increased. Chlorophyll a concentration and abundance of bacteria, oligotrich ciliates and crustacean zooplankton did not respond clearly to the flood event. Although the flood event lasted only a few days it took more than 2 weeks for the planktonic community to return to its original composition.  相似文献

Acro-cardio-facial syndrome (ACFS) is a rare genetic disorder characterized by split-hand/split-foot malformation (SHFM), facial anomalies, cleft lip/palate, congenital heart defect (CHD), genital anomalies, and mental retardation. Up to now, 9 patients have been described, and most of the reported cases were not surviving the first days or months of age. The spectrum of defects occurring in ACFS is wide, and both interindividual variability and clinical differences among sibs have been reported. The diagnosis is based on clinical criteria, since the genetic mechanism underlying ACFS is still unknown. The differential diagnosis includes other disorders with ectrodactyly, and clefting conditions associated with genital anomalies and heart defects. An autosomal recessive pattern of inheritance has been suggested, based on parental consanguinity and disease's recurrence in sibs in some families. The more appropriate recurrence risk of transmitting the disease for the parents of an affected child seems to be up to one in four. Management of affected patients includes treatment of cardiac, respiratory, and feeding problems by neonatal pediatricians and other specialists. Prognosis of ACFS is poor.  相似文献

Replication of herpes simplex virus takes place in the cell nucleus and is carried out by a replisome composed of six viral proteins: the UL30-UL42 DNA polymerase, the UL5-UL8-UL52 helicase-primase, and the UL29 single-stranded DNA-binding protein ICP8. The replisome is loaded on origins of replication by the UL9 initiator origin-binding protein. Virus replication is intimately coupled to recombination and repair, often performed by cellular proteins. Here, we review new significant developments: the three-dimensional structures for the DNA polymerase, the polymerase accessory factor, and the single-stranded DNA-binding protein; the reconstitution of a functional replisome in vitro; the elucidation of the mechanism for activation of origins of DNA replication; the identification of cellular proteins actively involved in or responding to viral DNA replication; and the elucidation of requirements for formation of replication foci in the nucleus and effects on protein localization.  相似文献
